Omeprazole is a proton pump inhibitor used to treat diseases such as peptic ulcer and gastroesophageal reflux disease (GERD). It can specifically act on the site of the proton pump in the gastric parietal cells and convert it into the active form of sulfenamide. It then irreversibly binds to the sulfhydryl group of the proton pump through a disulfide bond to form a complex of sulfenamide and the proton pump (H+-K+-ATPase), thereby inhibiting the activity of the enzyme, preventing the H+ in the parietal cells from being transported to the gastric cavity, blocking the last step of gastric acid secretion, and greatly reducing the amount of gastric acid in gastric juice. Therefore, this product has a strong and lasting inhibitory effect on gastric acid secretion caused by various reasons (such as basal gastric acid secretion and gastric acid secretion caused by histamine, pentagastrin and stimulation of the vagus nerve, including gastric acid secretion caused by dibutyl cyclic adenosine monophosphate that cannot be inhibited by H2 receptor blockers). This is related to the irreversibility of the inhibitory effect of this product on the proton pump. The acid secretion can only be restored after a new proton pump is formed.
